Abstract In recent years, there has been an increased need for new tools for chemical or enzymatic ligation that allow different protein or peptide fragments to be coupled in a very specific way. Progress has been made by using pure chemical approaches such as native chemical and Staudinger ligations. In parallel, the arsenal of biochemical ligation strategies, including expressed protein and enzyme ligations, has been enriched with novel enzymes such as V8 protease and staphylococcal sortase.
